    Yeasts are unicellular haploid fungi that help us to bake bread and ferment alcoholic beverages, but in some cases, they can cause infections that are sometimes fatal. Over 1000 different known species of yeasts are widely distributed in nature[1]. Although the majority of these (e.g., Saccharomyces cerevisiae) live in the general environment, a few species (e.g., Candida) are associated with humans and sometimes become pathogenic[1]. This occurs when our immune system weakens and Candida yeasts start to grow in an uncontrolled way, thereby causing candidiasis and nosocomial infections that result in high patient morbidity and mortality rates[2]

    Accommodating Employees With and Without Disabilities

    Efforts to recruit and retain employees with disabilities are often tempered by employers’ concerns over potential workplace accommodation costs. This study reports on accommodations requested and granted in intensive case studies of eight companies, based on more than 5,000 employee and manager surveys, and interviews and focus groups with 128 managers and employees with disabilities. Two unique contributions are that we analyze accommodations for employees without disabilities as well as for those with disabilities, and compare perspectives on accommodation costs and benefits among employees, their coworkers, and their managers. We find people with disabilities are more likely than those without disabilities to request accommodations, but the types of accommodations requested and the reported costs and benefits are similar for disability and non-disability accommodations. In particular, fears of high accommodation costs and negative reactions of coworkers are not realized; all groups tend to report generally positive coworker reactions. Multilevel models indicate granting accommodations has positive spillover effects on attitudes of coworkers, as well as a positive effect on attitudes of requesting employees, but only when coworkers are supportive. Consistent with recent theorizing and other studies, our results suggest the benefits from a corporate culture of flexibility and attention to the individualized needs of employees

    Progesterone significantly enhances the mobility of boar spermatozoa

    Progesterone released from the cumulus cells of the oocyte causes a number of physiological responses in human sperm cells including hyperactivation, acrosome reaction and chemotaxis. We employed a validated sperm mobility assay, which involves measuring the ability of sperm to penetrate an inert cell separation solution over time, to assess the ability of progesterone to enhance the mobility of boar spermatozoa. Cells maximally penetrate the solution over 50 minutes. 100nM progesterone significantly (P = 0.01) increased the mobility of non-capacitated sperm cells causing a doubling in the rate at which the cells penetrated through the cell separation solution (control half maximal penetration rate [Km] = 18.0±2.2; +100nM progesterone Km = 8.8±0.8min). Similarly, capacitated cells penetrated at a rate (Km = 19.2±3.0 min) not significantly different from non-capacitated cells and 100nM progesterone also significantly increased the rate of penetration of capacitated cells (Km = 9.5±1.0 min, P<0.05). The T-type voltage gated calcium channel blocker mibefradil (30mM) significantly inhibited both the control and progesterone enhanced mobility in non-capacitated and capacitated sperm. Only capacitated cells showed a significant increase in the acrosome reaction in response to 100nM progesterone (control non-reacted = 75±4%, +100nM progesterone non-reacted = 47±10%). Western blot analysis confirmed that there was an increase in the total protein tyrosine phosphorylation levels in capacitated cells. In conclusion, we have demonstrated that 100nM progesterone accelerates the mobility of boar sperm cells through an inert cell separation solution in an extracellular calcium dependent manner

    Host carbon sources modulate cell wall architecture, drug resistance and virulence in a fungal pathogen

    The survival of all microbes depends upon their ability to respond to environmental challenges. To establish infection, pathogens such as Candida albicans must mount effective stress responses to counter host defences while adapting to dynamic changes in nutrient status within host niches. Studies of C. albicans stress adaptation have generally been performed on glucose-grown cells, leaving the effects of alternative carbon sources upon stress resistance largely unexplored. We have shown that growth on alternative carbon sources, such as lactate, strongly influence the resistance of C. albicans to antifungal drugs, osmotic and cell wall stresses. Similar trends were observed in clinical isolates and other pathogenic Candida species. The increased stress resistance of C. albicans was not dependent on key stress (Hog1) and cell integrity (Mkc1) signalling pathways. Instead, increased stress resistance was promoted by major changes in the architecture and biophysical properties of the cell wall. Glucose- and lactate-grown cells displayed significant differences in cell wall mass, ultrastructure, elasticity and adhesion. Changes in carbon source also altered the virulence of C. albicans in models of systemic candidiasis and vaginitis, confirming the importance of alternative carbon sources within host niches during C. albicans infection

    Investigations on RF Behavior of a V-Band Second Harmonic Gyrotron for 100/200 kW Operation

    This article presents the investigations on RF-behavioral aspects for the possible operation of a V -band, continuous wave (CW) second harmonic gyrotron for plasma diagnostic application. Keeping in view the design goals and constraints, initial design studies for the mode selection and the computation of starting currents are carried out. From these studies, two possible modes, namely, TE 7,3 and TE 8,3 are considered for the second harmonic operation. Later, the cold cavity design and self-consistent calculations are carried out for the selected operating modes. All the computations are performed using the latest version of our in-house code Gyrotron Design Studio Second Harmonic Version 2020 (GDS2H-2020) with Glidcop as the cavity material. The RF behavior studies confirm the feasible operation of such a second harmonic gyrotron with power levels in excess of 115.52/217.64 kW with the chosen modes of operation

    Passive-performance, analysis, and upgrades of a 1-ton seismic attenuation system

    The 10m Prototype facility at the Albert-Einstein-Institute (AEI) in Hanover, Germany, employs three large seismic attenuation systems to reduce mechanical motion. The AEI Seismic-Attenuation-System (AEI-SAS) uses mechanical anti-springs in order to achieve resonance frequencies below 0.5Hz. This system provides passive isolation from ground motion by a factor of about 400 in the horizontal direction at 4Hz and in the vertical direction at 9Hz. The presented isolation performance is measured under vacuum conditions using a combination of commercial and custom-made inertial sensors. Detailed analysis of this performance led to the design and implementation of tuned dampers to mitigate the effect of the unavoidable higher order modes of the system. These dampers reduce RMS motion substantially in the frequency range between 10 and 100Hz in 6 degrees of freedom. The results presented here demonstrate that the AEI-SAS provides substantial passive isolation at all the fundamental mirror-suspension resonances

    Huddle test measurement of a near Johnson noise limited geophone

    In this paper, the sensor noise of two geophone configurations (L-22D and L-4C geophones from Sercel with custom built amplifiers) was measured by performing two huddle tests. It is shown that the accuracy of the results can be significantly improved by performing the huddle test in a seismically quiet environment and by using a large number of reference sensors to remove the seismic foreground signal from the data. Using these two techniques, the measured sensor noise of the two geophone configurations matched the calculated predictions remarkably well in the bandwidth of interest (0.01 Hz–100 Hz). Low noise operational amplifiers OPA188 were utilized to amplify the L-4C geophone to give a sensor that was characterized to be near Johnson noise limited in the bandwidth of interest with a noise value of 10−11 m/Hz⎯⎯⎯⎯⎯√10−11 m/Hz at 1 Hz

    Rule-Based Forecasting: Using Judgment in Time-Series Extrapolation

    Rule-Based Forecasting (RBF) is an expert system that uses judgment to develop and apply rules for combining extrapolations. The judgment comes from two sources, forecasting expertise and domain knowledge. Forecasting expertise is based on more than a half century of research. Domain knowledge is obtained in a structured way; one example of domain knowledge is managers= expectations about trends, which we call “causal forces.” Time series are described in terms of 28 conditions, which are used to assign weights to extrapolations. Empirical results on multiple sets of time series show that RBF produces more accurate forecasts than those from traditional extrapolation methods or equal-weights combined extrapolations. RBF is most useful when it is based on good domain knowledge, the domain knowledge is important, the series is well behaved (such that patterns can be identified), there is a strong trend in the data, and the forecast horizon is long. Under ideal conditions, the error for RBF’s forecasts were one-third less than those for equal-weights combining. When these conditions are absent, RBF neither improves nor harms forecast accuracy. Some of RBF’s rules can be used with traditional extrapolation procedures. In a series of studies, rules based on causal forces improved the selection of forecasting methods, the structuring of time series, and the assessment of prediction intervals

    Environmental Noise in Advanced LIGO Detectors

    The sensitivity of the advanced LIGO detectors to gravitational waves can be affected by environmental disturbances external to the detectors themselves. Since the transition from the former initial LIGO phase, many improvements have been made to the equipment and techniques used to investigate these environmental effects. These methods have aided in tracking down and mitigating noise sources throughout the first three observing runs of the advanced detector era, keeping the ambient contribution of environmental noise below the background noise levels of the detectors. In this paper we describe the methods used and how they have led to the mitigation of noise sources, the role that environmental monitoring has played in the validation of gravitational wave events, and plans for future observing runs
